Rola układu glutaminergicznego w patomechanizmie chorob afektywnych i mechanizmie działania nowych lekow przeciwdepresyjnych

W ostatnich latach zgromadzono liczne dowody wskazujące na rolę układu glutaminergicznego w patogenezie chorób afektywnych i mechanizmie działania leków przeciwdepresyjnych. Jednym z istotnych mechanizmów działania przeciwdepresyjnego jest osłabienie ekspresji i funkcji receptora glutaminergicznego NMDA. W ostatniej dekadzie uzyskano dowody na możliwość uzyskania szybkiego efektu terapeutycznego w depresji poprzez wpływ na układ glutaminergiczny stosowania procedury jednorazowego wlewu antagonisty receptorów NMDA – ketaminy. Oprócz ketaminy przedstawiono dane wskazujące na możliwość działania przeciwdepresyjnego innych środków wpływających na układ glutaminergiczny, takich jak: D-cykloseryna, amantadyna, memantyna, traksoprodil, riluzol oraz jony cynku i magnezu. Dalsze przybliżenie roli układu glutaminergicznego w mechanizmie działania przeciwdepresyjnego może stwarzać nowe perspektywy terapeutyczne w chorobach afektywnych.

The role of glutamatergic system in the pathogenesis and mechanism of action of novel antidepressants

In recent years, an increasing evidence has been accumulated pointing on a role of glutamatergic system in the pathogenesis of mood disorders and in the mechanism of action of antidepressants. The one of the most important mechanism of antidepressant action is reduction of the expression and function of glutamatergic receptor NMDA. In recent decade an evidence has been obtained for a rapid antidepressant effect connected with the influencing glutamatergic system by single infusion of NMDA antagonist receptor, ketamine. Besides of ketamine, data are provided concerning a possibility of antidepressant action for other substances influencing glutamatergic system such as D-cycloserine, amantadine, memantine, traxoprodil, riluzole as well as zinc and magnesium ions. Further elucidation of a role of glutamatergic system in the mechanisms of antidepressants may provide new therapeutic perspectives in mood disorders.
