Nowe spojrzenie na ceruloplazminę

Ceruloplazmina (Cp) jest enzymem syntetyzowanym głównie w wątrobie, pełniącym różnorodne funkcje w organizmie. Bierze udział nie tylko w transporcie miedzi do tkanek oraz utrzymaniu jej na odpowiednim poziomie w organizmie, ale również uczestniczy w metabolizmie żelaza. Jako ferrooksydaza utlenia jony Fe(II) do Fe(III), umożliwiając ich przyłączenie do białek transportujących (transferyna) lub magazynujących (ferrytyna). Ponadto Cp jest białkiem ostrej fazy, reguluje poziom amin biogennych (adrenalina i noradrenalina) w organizmie, a także wykazuje aktywność antyoksydacyjną i prooksydacyjną. Zaburzenia biosyntezy ceruloplazminy spowodowane mutacjami genu kodującego to białko prowadzą do zakłócenia metabolizmu żelaza i gromadzenia tego pierwiastka w różnych narządach (szczególnie mózgu), skutkiem czego jest stres oksydacyjny powodujący poważne uszkodzenia neuronów. Niskie stężenie Cp w surowicy i związane z tym odkładanie żelaza w organizmie sprzyja powstawaniu i rozwojowi wielu chorób neurodegeneracyjnych (aceruloplazminemia, choroba Wilsona, Parkinsona i Alzheimera).

A new look on ceruloplasmin

Ceruloplasmin (Cp) is an enzyme synthesized primarily in the liver, which performs a variety functions in our body. It not only participates in the transport of copper to tissues and maintaining it in the correct level but also take parts in the metabolism of iron. As a ferroxidase, oxidizes Fe(II) to Fe(III), allowing their attachment to transport (transferrin) or storing proteins (ferritin). Furthermore, Cp is an acute phase protein, regulates the level of biogenic amines (adrenaline and noradrenaline) and has antioxidative and prooxidative activities. Disorders in biosynthesis of ceruloplasmin caused by mutations in the gene encoding this protein leads to disruption of iron metabolism and accumulation of this element in various organs, (especially in the brain). This induce the oxidative stress and causes serious damage in the neurons. Low concentration Cp in the serum and associated with it deposition of iron in the body, promotes the formation and the development of many neurodegenerative diseases (aceruloplasminemia, Wilson’s, Menkes and Alzheimer’s diseases).
