Transportery leków w łożysku

Łożysko jest jedynym narządem regulującym wymianę między matką a płodem, funkcjonując jako bariera półprzepuszczalna. Białka błonowe zaangażowane w transport substancji pochodzenia endogennego oraz ksenobiotyków występują w łożysku ludzkim, w syncytiotrofoblaście oraz śródbłonku naczyń płodowych. Najistotniejszymi wśród nich są transportery nadrodziny ABC: glikoproteina P, BCRP i MRP, które determinują przenikanie wielu leków przez barierę łożyskową, a tym samym bezpieczeństwo farmakoterapii. W opracowaniu tym, przedstawiamy przegląd wiedzy na temat barierowej roli transporterów w łożysku ludzkim.

Drug transporters in placenta

Placenta is a crucial organ that regulates exchange of endo- and exogenous compounds between mother and fetus, functioning as a semi-permeable barrier. Membrane proteins involved in the transport of substances and xenobiotics are expressed in human placenta, and are localized in syncytiotrophoblast and fetal vascular endothelium. The most important transporters expressed in the placenta belong to the superfamily of ABC transporters: P-glycoprotein, MRPs and BCRP. The transporters determine penetration of many drugs through the placenta, and thus the safety of pharmacotherapy. In this study, we present an overview of the barrier role of transporters in the human placenta.
