2,4-thiazolidinediones as peroxisome proliferator-activated receptor PPARg agonists with anticancer properties

2,4-Thiazolidinediones, otherwise known as glitazones, are synthetic PPARγ receptor agonists with antidiabetic, antiinflammatory and antitumor activity. Since the 1990s, compounds such as pioglitazone and rosiglitazone have been successfully used in the treatment of type II diabetes and are currently being tested in new indications. PPARγ receptors are transcription factors that belong to the family of hormonal nuclear receptors. Their main function is connected with regulation of fatty acid metabolism and reduction of insulin resistance of peripheral tissues. Recent reports also indicate the participation of these receptors in inhibiting of carcinogenesis. Activation of PPARγ causes apoptosis of cancer cells, inhibition of their proliferation and angiogenesis with inducing of normal cell differentiation. In this paper general information about the family of PPAR receptors and their synthetic ligands – thiazolidinediones was presented with emphasis on their antineoplastic activity.
