Solid phase microextraction (SPME) as a modern technique for the isolation of medications from biological samples

Laboratory analysis of exogenous compounds (e.g., medicinal substances) as well as endogenous ones (e.g., biomarkers) is a multistage process, requiring the use of advanced analytical methods that facilitate qualitative and quantitative determination of the level of a given compound in the analyzed sample. This in turn allows for the calculation of a concentration of analyte in the body, and in the case of medications – estimation of the correct dosage, drug metabolism and the risk of side effects. Modern extraction techniques of drugs from the biological matrix aim at the miniaturization of the entire procedure in order to limit the consumption of toxic compounds and reduce the amount of biological material collected from the patient. One of the modern microextraction techniques is solid-phase microextraction (SPME). It allows the extraction of even trace amounts of compounds from conventional (e.g., blood) as well as unconventional (e.g., saliva) biological matrices. Due to the simplicity and speed of SPME extraction, this technique becomes increasingly popular in pharmaceutical analysis, and also in forensic and clinical studies.
